Are you talking about a site map, or an actual map of something (like a map of a real estate development or an amusement park for example). Can you give a little more detail about the purpose of the map?

You might consider a zoom feature, so that people can see the location of the company within the context of the rest of the area, as well as a close up of the "neighborhood" with relevant "landmarks". If the company has a huge building or complex (with multiple entrances and/or parking areas) you might add a 3rd zoom level on just that, with some labels.

I would definitely add a hover feature which pops up more info, like you said. Whether you make the hot spots with 3D buildings or not is really an aesthetics thing -- it really depends on the map; in some cases it might end up looking cluttered, which of course would make it difficult to read.

I also recommend having some static text with the full address of the company too. And it's a good idea to have a "printable version" available so that people can take the map with them in the car.
